Q: What if teacher Manaia doesn’t accept our current project?

A: Don’t worry about that as we can do nothing about it, even if she doesn’t accept it, we still
have the option to change again and appoint another presentation

(UPDATE FOR QUESTION 1):

Ignore question 1 she already answered.

Q: What do we have to do to the presentation tomorrow, we have nothing are we fucked?

A: No because most groups only started today working something out and the presentation
tomorrow is just to expose the topics we are working on to the teachers (you don’t have to
know the whole process of the project yet)

Topics for the presentation:

Topic 1:

Explanation and exposition of the polymer present on the plastics we want to degrade.

Topic 2:

Exposition of the enzymes used for degradation of the said polymer present in plastics that
isn’t biodegradable

Topic 3:

Explaining how to synthesize and produce the enzyme capable of completing our objective
(probably bigger topic than the others maybe requires 2 people)

Topic 1:

Polyethylene terephthalate (PET), is a thermoplastic polymer resin which isn’t biodegradable


and is used in plastic products, since it isn’t biodegradable, the accumulation of this polymer
poses serious environmental threats, hence the pollutant designation of this polymer. This
obviously also means that the accumulation of this synthetic material is a serious threat to
ecosystems and biodiversity. The main objective of this project is to find a way to degrade said
polymer in order to remove it from the environment.

Topic 2:

In order to accomplish the problem posed in topic 1 there has been discovered a bacterial
strain (Ideonella sakaiensis 201-F6) that produces unique enzymes capable of degrading said
polymer (depolymerize). These enzymes are polyethylene terephthalate hydrolase (PETase)
and mono(2-hydroxyethyl) terephthalic acid hydrolase, these enzymes work together to
depolymerize the base polymer. The objective is to determine the structure of these enzymes
and recreate their function in order to learn more about this whole process and to produce
some kind of benefit with this project, as well as optimizing the function of the scoped
enzymes.

Topic 3:

Identifying the main sequence of Poly(ethylene terephthalate) hydrolase amino acids (bacterial
based) in order to insert it in a plasmid for future lab protein synthesis recreation, for future
use and experimentation (NEED TO ADD MORE TO THIS TOPIC)

Topic 4:

To sum it up this would be a project we would be very happy to deliver as it delves in genetic
manipulation which is a highly interesting area of research for all the elements present in the
group??, and we would very much like to accomplish a general benefit with the conclusion of
this project.
