Buffer Solutions Preparation Guide

Buffer Solutions Composition:-

A. Acetate Buffer (0.2 mol/liter):-


Place x ml of 0.2 mol/liter acetic acid in a 100 ml volumetric flask and make up top the mark with
0.2 mol/liter sodium acetate.

B. Carbonate/ Bicarbonate Buffers (0.1 mol/liter):-


Place x ml of 0.1 mol/liter Sodium Bicarbonate into a 100 ml volumetric flask and make up top the
mark with 0.2 mol/liter Sodium Carbonate.

C. Citric Acid/ Sodium Citrate Buffers(0.05 mol/liter):-


Place x ml of 0.05 mol/liter Citric acid in a volumetric flask and make up top the mark with 0.05
mol/liter Trisodium citrate.

D. Phosphate Buffers (0.1 mol/liter):-


a. Sodium Phosphate Buffers:-
Place x ml of 0.2 mol/liter Sodium Hydroxide to 50 ml of 0.2 mol/liter Sodium dihydrogen
phosphate and dilute to 100 ml.

b. Potassium Phosphate Buffer:-


Place x ml of 0.2 mol/liter Potassium Hydroxide to 50 ml of 0.2 mol/liter Potassium dihydrogen
phosphate and dilute to 100 ml.

E. TRIS (Hydroxymethyl) Amino Methane/ HCL Buffer (0.1mol/liter):-


Place x ml of 0.2mol/liter Hydrochloric acid to 50 ml of 0.2 mol/liter tris and make up to 100 ml
with water.

F. Barbitone Buffer (0.07 mol/liter, pH 8.6):-


Dissolve 2.58g of diethylbarbituric acid and 14.42g of sodium diethylbarbiturate in water and make
up to 1 liter.

G. Cacodylate Buffer (0.1 mol/ liter, pH 6.5):-


Add 29 ml of 0.2 mol/liter HCl to 50 ml of Sodium Cacodylate and make up to 100 ml with water.

H. Cacodylate Buffer (0.1 mol/ liter, pH 6.5):-


Add 16 ml of 0.2 mol/liter HCl to 50 ml of Sodium Cacodylate and make up to 100 ml with water.

I. Phthalate Buffer (0.1 mol/liter, pH 6.0):-


Add 43 ml of 0.1 mol/liter NaOH to 50 ml of 0.2 mol/liter potassium hydrogen phthalate.
