Rheumatoid factor is an antibody that is present in the blood of some individuals with rheumatoid arthritis but not in healthy individuals. The rheumatoid factor test detects the presence of this antibody to help diagnose rheumatoid arthritis. The test involves mixing patient serum with latex particles coated with human proteins. If rheumatoid factor is present, it will cause the latex particles to agglutinate or clump together. A positive result indicates the level of rheumatoid factor is above 8 IU/ml. The test is qualitative to detect presence or absence of rheumatoid factor and can also be semi-quantitative to determine the antibody titer level. False positives can occasionally occur so the test result should be considered along with clinical examination.

Rheumatoid Factor

Introduction
Rheumatoid factor is an antibody that is measurable in the blood with a
routine blood test. Rheumatoid factor is actually an antibody that can bind to
other antibodies. Antibodies are normal proteins in our blood that are
important parts of our immune system. Rheumatoid factor is an antibody that
is not usually present in the normal individual. Because rheumatoid factor
antibody binds to normal antibodies, it can be generally referred to as an
autoantibody. Health-care professionals use the rheumatoid factor test to
assist in the diagnosis of rheumatoid arthritis

Principle
detection of Rheumatoid factor by using Latex Agglutination
Purpose ,RF-latex is a slide agglutination test for the qualitative and
semiquantitative detection of RF in human serum. Latex particles
coated with human gammaglobulin are agglutinated when mixed
with samples containing RF.

Procedure
1. the reagents and samples are reached to room temperature. The
sensitivity of the test may be reduced at low temperatures.
2. Place 50 L of the sample and one drop of each Positive and
Negative controls into separate circles on the slide test.
3. Swirl the RF-latex reagent gently before using and add one drop
(50 L) next to the sample to be tested.
4. Mix the drops with a stirrer, spreading them over the entire surface
of the circle.

*for Semi-quantitative method


serial two fold dilutions of the sample in 9 g/L saline solution is made each dilutions Proceed for as in the
qualitative method

Result
In quantitative test : agglutination was formed this mean the
result was positive . ( antibody > 8 IU/ ml )
In semi-quantitative test ;
1 :3 .. positive
1:9 .. positive
1 :27 . Negative
Titer = 9*8 = 72 IU/ml

Discussion
INTERFERENCES Hemoglobin (up to 10 g/L), bilirubin (up to
20 mg/dL) and lipemia (up to 10 g/L) do not interfere. Other
substances may interfere6. LIMITATIONS OF PROCEDURE The incidence of false positive results is about 3-5 %.
Individuals suff ering from infectious mononucleosis,
hepatitis, syphilis as well as elderly people may give
positive results. - Diagnosis should not be solely based on
the results of latex method but also should be
complemented with a Waaler Rose test along with the
clinical examination
