MICROBIOLOGY AND PARASITORY FOR ENVIRONMENTAL
ENGINEERING
LABORATORY ACTIVITY #1
Title: The Microscope
Introduction:
The microscope is the ost important instrument used by the microbiologist
and bacteriologist, and it’s care and correct usage are vital
To the successful technical skills. Any difficulty encountered should
instantly brought to the attention of the instructor. It is the function
Of the microscope to make visible those things that are minute or very
small things to be seen by the naked eye. This is being done by
A system of lenses of sufficient magnification and resolving power, so that
the small elements which are close together in specimen
Appear to be distinctly separated, thus giving a clearly defined image,
when sufficient illumination is used. The total magnification of the
Microscope is the product of the magnification due to the objective and the
magnification due to the eyepiece.
I. OBJECTIVE:
1. To identify the parts of the microscope and their functions.
2. To become familiar with the variations of light microscopy.
3. To learn how to use the Microscope effectively.
4. To manipulate the microscope..
II. APPARATUS/ Materials:
Compound Microscope, Glass slide,Any specimen to be viewed and
reference material
III. PROCEDURES:
1. Operation of the Microscope
a) Place the microscope in proper working table for proper viewing.
b) Locate all parts of the microscope.
2nd Semester, SY 2019-20 Page 1 of 3
c) Use the reference material for microscope.
2. Safety Precaution
a) Prevention of infection
b) Installation of the device
c) Electrical safety
d) LED (light emmiting diode)
e) Safety symbols
3. Intended Use
It is designed to be used to observe magnified images of specimen in
various routine work and research
Application. This include observation of living celss or specimen
taken from tissues to gain physiological
Or morphological information at the laboratories.
4. Handling Precaution
a) The microscope is a precision instrument, handle it with care
and avoid subjecting it to a sudden
Or severe impact.
b) Never dissemble any parts of the product, failure could be
caused by it.
c) When carrying the microscope be careful not to touch the
objectives.
5. Maintenance and storage
a) Clean the microscope when the experiment ended.
b) Leave the microscope with the lowest power objective in
working position.
c) When not in use store in a dry place or cover it with dust proof
cover.
IV. ILLUSTRATION:
1. DRAW and Label the Parts of the Microscope ( it can be Printed)
V. QUESTION FOR RESEARCH
1. Define the following terms:
a) Bifocal
b) Contrast
c) Parfocal
d) Resolution
e) Magnification
