This document provides a monograph for amprolium hydrochloride, including its structure, uses, definitions, characteristics, identification tests, and specifications. Amprolium hydrochloride is an antiprotozoal drug used to prevent and treat coccidiosis in veterinary medicine. It is a white or colorless powder that is freely soluble in water and slightly soluble in ethanol. Identification tests include infrared spectroscopy, UV spectroscopy, and chemical tests. Purity tests specify limits for picoline and loss on drying of less than 1.0% of weight and sulfated ash of less than 0.1%.

Topic:
Monograph
(Amprolium hyclrochloride)

Table of content:
1. Structure

2. Action and use

3. Defination

4. Characteristics

5. Identifigation

6. Tests
7. Loss on drying
8. Sulphate ash
 Structure:
 Amprolium Hydrochloride:

 Action and use:

Antiprotozoal;prevention

and treatment of coccidiosis.(veterinary).

 Definition:
Amprolium hydrochloride

is 1-(4-amino-2-propylpyrimidin-5-ylmethyl)-

2-methylpyridinium chloride hydrochloride.It

contain not less than 97.5%and not more

101.0% of C14H 19CIN4,HCL, calculated with

reference to the dried substance.

 Chracteristics:
A white or almost
white powder;odourless or almost
colourless.Freely soluble in water,slightly
soluble in ethanol(96%)very slightly soluble in
ether,practically insoluble in chlorojium.
 Identification:
A. The infrared absorbtion spectrum,appendix
2A is concordant with the refrence
spectrums of amprolium hydrochloride(RSV
07).
B.The light absorbtion, appendix 2b,is the
range 230 to 350 nm of 0.002% w/v solution
in 0.1M hydrochloric acid exhibits two
maxima, at 246nm and 262nm the
absorbances at the maximum are about 0.84
and about 0.80,respectively.
C.To 1 mg add 5ml of naphthalenediol regent
solution;a deep violet colour is produced.
D.Yields the reaction characteristics of
chlorides appendix VI.
 Tests:
 Picoline:
Dissolve 1.5g in 30 ml of water in a distillation
flask,add 20ml of a saturated solution of
potassium carbonate.Sesquihydrate,connect
the flask to a coarse-fritted aerator extending
to the bottom of a 100ml gradiated cylinder
containing 50ml of 0.05m hydrocloric acid and
uthrough sulphuric acid and glass
wool, through the system for 60minutes to
5ml of hydrochloric acid solution and
sufficient 0.05m hydrochloric acid to produce
200ml.The absorbance of the resulting
solution at 262 nm is not greater than
0.52,appendix 2b.
 Loss on drying .
When dried to
constant weight at 100 at a pressure not
exceeding 0.7kpa,losses not more than 1.0%
of it weight,Use 1 g.
 Sulphated ash:
Not more than
0.1%,appedix 1X A.
 Refrence:
British
pharmowpeia.
