PERMANGANATE VALUE (PV)


(BOD / COD & TOC)

The Permanganate Value is determining the general quality of Final Effluents in sewage.
Based on the results it will give the OK for the discharge of Sewage water.
The test procedure is relatively easy and consists of 4 steps:

Ÿ Three sample containers in the set must be filled with 100 ml of Sewage Effluent.
Ÿ Add two Acidifying SE tablets per sample container and shake it to mix well.
Ÿ Add one permanganate value tablet to the first sample container, then add two permanganate
tablets to the second and three tablets to the third, and again shake them well until it all of them
are mixed and dissolved.
Ÿ Wait 30 minutes! Then check how many tubes have remained pink and read the result from
the following table:

In the Test Kit you will also find instructions for the testing of Crude waste, the same is valid for Settled

BOD / COD & TOC

BOD = Biochemical Oxygen Demand


COD = Chemical Oxygen Demand
TOC = Total Organic Carbon

The results from this test are in connection with the Permanganate Value (PV) as done in paragraph
1. to convert the PV for domestic sewage and effluent to probable BOC, COD and TOC values multiply
by the following factors:

In general there is a connection between the Turbidity and the BOD-value of settled sewage and efluent.
The BOD can also be calculated from the result of the turbidity test usig the fomula:

BOD-values can be cross checked that way with the PV tests. Recommendation for Effluent is not more
than 20 mg/l.

Operation:

Make sure the tester is calibrated to pH 7.00


1) Remove protective cap and switch meter on.
2) Immerse electrode in solution to be tested.
3) Stir gently and wait for the reading to stabilize.
4) Press “°C/°F“ to switch between Celsius and Fahrenheit.
5) Read the result, than rinse the electrode with water.
6) Replace the protective cap and switch the meter off.
7) Make sure the electrode is kept wet all the time!

Weekly monitoring of bacteria levels has been recommended by many legislators and professional
authorities as a visual performance indicator to both system and treatment regime. This allows the
user to gauge how effective a chemical or biocide product is in the particular application and a trend
can be quickly established identifying changes and taking quick remedial action where required. It
should be noted from the outset that Dipslides alone do not detect Legionella as a select micro-
organism, however it is generally accepted that overall bacteria levels in excess of 10“ are considered
able to support Legionella and obviously a serious risk.

The dipslide consists of a plastic paddle with culture media on each side, the tube keeps the media
both moist & sterile until required. The product most suitable is a slide based on a standard nutrient
agar with a red dye supplement added during production, this has the advantage of showing any
viable colonies as red dots, easily identified and compared against the comparison chart.

IMPORTANT - Monitoring is not a substitute for a treatment regime, it is always best to seek the
advice of a professional water treatment or environmental company who will supply a risk

Dipslides
Application, Storage & Disposal

Storage
Dipslides should be stored in cool dry conditions, but not in a refrigerator - prior to despatch our
stock is stored in a cold room at 10'C in order to prevent condensation and de-hydration of the
agar and ensure Dipslides arrive in perfect condition.

Shelf Life
Dipslides have a typical use by date of 9 months dependent of manufacture cycle, they can be
used after this date as long as no contamination or visible shrinkage shows on the agar surface -
excess water in the bottom of the slide would indicate the storage temperature was too high.

Application
Dipslides can be used to monitor microbial growth wherever the potential may exceed 1000 (101)
organisms per ml of sample fluid.

Industrial Waters: For detection of slime forming bacteria in cooling & industrial
waters, storage tanks and for evaluating biocide perfomiance in treated systems.

Industrial Fluids: For detection of bacteria & moulds in metal-working fluids, paper
processing waters, fuel tanks etc.

Environmental Hygiene: To monitor the surface contamination within domestic or


institutional sites.

Leather Industry: For detection of spoilage organisms in hide and skin soaking
liquors.

Disposal of Used Slides


Contaminated slides should be sterilised before disposal by immersing in a disinfectant or by
autoclave or incineration. After soaking in a disinfectant for 24 hours the dips can be disposed of
in any normal non hazardous waste service.

Dipslides are manufactured in a clean environment and stored at constant temperature prior to

The PDM/MAC dipslide allows convenient enumeration of Pseudomonas spp. on the pale straw
Pseudomonas differential medie (PDM) and enterobacteriaceae on the reddish/brown MacConkey
side. PDM agar is a differential media that selects for Pseudomonas spp, while MacConkey agar is
differential, all in one test for gram negative bile tolerant enterobacteriaceae.

SAMLLING: AIR
The sample should be taken by inverting the paddle and inserting in the vial. Let it sit for 15 minutes

SAMPLING: FLUIDS
The sample should be taken by immersing both sides of the paddle into the fluid to be tested, it having
first been removed from the sterile container. Excess sample should be gently shaken from the paddle

The sample should be taken by allowing direct contact between te agar surface and the test material.
The paddle is flexible and can be bent at the upper end to allow both surfaces to come into intimate
contact. Bacterial recovery rate is about 50% so that sweeping an area approximately twice that of the
paddle will give a more accurate result.

INCUBATION:
Incubate at 30° C for 24-48 hours, when full enumeration should be completed.

DISPOSAL
Used slides should be incinerated or autoclaved. Alternatively, immerse in a 10% bleach solution for
24 hours.

How to properly dilute samples to extend measurement range


In some cases, the value of your water sample exceeds the maximum measurement range of the
method you choose to test.
Example: Expected content of CHLORIDE in your water sample is 1200 ppm whilst „method ID 10"
on PrimeLab only allows to test the range 0 - 25 ppm.

To still be able to test your water sample, a proper dilution can be performed.
The following steps describe how to do:

IMPORTANT:
• Dilution to extend the measurement range does not apply to titration methods (MINI-Kits) but
only to Photometer (PrimeLab) measurements
• In best case, use lab pipettes (variable 1-10ml -PLSp-PIP10ml- and 0.1 - 1ml -PLSp-PIP1ml-) and
always use new (never used!) pipette tips
• Always use deionized / destilled water for dilution
• Do use a clean tube (100ml shaker/dislution tube, for example) to mix

PROCEDURE:
1) Homogenousely mix your sample water with deionized/destilled water according to the table below.
2) Use 10ml of your mixture for testing
3) Once a result is displayed, multiply the result by factor according to table below
4) If you use PrimeLab software and/or app, manually change synchronized result to the real value
(reading x factor) and add a comment to the comment field, such as „dilution with factor...“

If you use „Extend range by factor 10" and PrimeLab displays „12 ppm“ as a result, your real
result is 120 ppm (factor 10)

NOTE:
With each dilution step, accuracy goes down as slight tolerances to the amount of water taken apply,
even when using lab-pipettes!

FILTRATION
BOILER WATER

While conducting water testing, there are cases in which collected samples
< 28 Bar may be turbid. This complicates the end point color & the results may not be
28 - 56 Bar correct. In such cases, filtration of the turbid sample with a special filter paper
Is necessary.

Oxygen Scavengers Boiler: From Boiler Blowdown, as instructed by the manufacturer. Sample
should be cooled to < 26° C.
Sampling Procedure: Flush sampling line a few minutes then reduce the flow
for 3-5 minutes with no disturbance. Flush the sample bottles a few times with
the sample water. Collect the sample in the bottle w/o touching the sampling
line. If the analysis is to be delayed, sample must be kept in tightly closed
sample bottles.

Feedwater: From main feedline (as close as possible the boiler).

5) Crush the tablet with a clean stirrer until it has completely dissolved.

sampling chamber of the PrimeLab. Align the arrow on the front of the cuvette.

7) After the lapse of a 0:25 minute countdown the determined result is
displayed.

After pressing the button 4 (OK) the measurement is continued to determine the

! total chlorine (tCl) and later also bonded chlorine (cCl). If this is not required the
measurement can be termianted by pressing "HOME". IN this case only the value
for free chlorine (fCl) is stored.

8) Remove the lid again and add a "DPD N° 3 Photometer" tablet into the
same sample you have just used for determining "free chlorine".

9) Crush the tablet with a clean stirrer until it has completely dissolved.

10) Replace the lid on the cuvette, swirl it carefully and insert it into the
sampling chamber of the PrimeLab. Align the arrow on the front of the
4 cuvette.
Press TEST.

11) After the lapse of a 2 minute countdown the total result is displayed,
2 divided in
3 "fCl" = "free chlorine“
"cCl" = "combined chlorine“
"tCl" = "total chlorine".
4 Scroll through the results using the buttons "arrow up" and "arrow down".
Press button 4 to save the result.

Note(s)

-> Sample water with a high calcium content or high conductivity will cloud the
sample and deteriorate the measurement precision. In this case use the
DPD N° 1 High Calcium (HC) and DPD N° 3 High Calcium (HC) tablets.

-> If the measuring water contains further reducing chemicals (e.g. active
oxygen, bromine etc.) this will also be detected and is part of the result.

-> When preparing the measurement procedure it is important to avoid any


chlorine escaping, which can happen during pipetting and shaking the
sample. The measurement should be performed directly after sampling.

-> It is important that the measurement devices to be used have not been
cleaned with household detergent, as this could greatly reduce the
measurement. To prevent any contamination the cuvette, the cuvette lid and
the stirrer should be stored in a 0.1 g/l sodium hypochlorite solution for one
hour and then thoroughly rinse with distilled water.

-> The DPD reagent used in this procedure buffers the pH value of the sample
water in the range between 6.2 and 6.5 pH. If your sample water is very
alkaline or acidic this must be adjusted to a pH range between 6 and 7 by
the addition of 0.5 mol/l sulphuric acid or resp. 1 mol/l caustic soda before
the DPD reagent is added.

-> Water samples with parameter levels ´higher´ than the defined range may
lead to errors with the DPD chemistry; resulting in an incorrect reading
(possibly showing none detected). For measurement of higher chlorine
values please select the respective matching procedure.

10) Replace the lid on the cuvette and swirl it carefully to mix the solution

sure the arrow on the front of the cuvette is aligned. Press the TEST

-> For the analysis of total copper the following procedure is necessary:

- Add concentrated sulfuric acid to the test sample (1 ml per 100 ml of test
sample). By boiling it for 10 minutes everything is dissolved. Now cool the
test sample. Then add ammonia and bring the sample to a pH value of 3 – 5.
The initial volume of 100 ml of fluid have to be filled up with deionized water.
The analysis can now be performed as described with 10 ml of the liquid
obtained.

- With organic compounds pretreated water may need to be oxidized


(destruction of the copper complexes). Add concentrated sulfuric acid and
concentrated nitric acid to the test sample (1 ml per 100ml each). Now cool
the test sample. The analysis can now be performed as described.

-> For the analysis the water has to have a pH value of 4 - 6. Strongly acidic
water having a pH value of <2 should be neutralized with 8 mol per litre
potassium hydroxide KOH.

-> Not yet completely dissolved powder has no effect on the accuracy of the
measurement.

-> Disorders:

- Cyanide CN: To ensure full color development, the test sample had to be
enriched with 0.2 ml of formaldehyde and wait 4 minutes. The analysis can
now be performed as described. The test result must be multiplied by 1.02.

- Silver Ag +: Silver can cause blackening of the test sample. Add saturated
potassium (10 drops per 75 ml). Then the test sample had to be poured
through a fine filter. The analysis is now carried out as described with 10 ml
of the filtered liquid.

1) Pour 500 ml of sample water into a blender and mix at the highest
speed for 2 minutes.

One-Time-Zero: If after switching on the device you have already performed the

2) Fill 10ml distilled water into a clean 24mm cuvette.

3) Replace the screw lid on the cuvette and insert it into the PrimeLab
4 sampling chamber. Align the arrow on the front of the cuvette. Press
ZERO.

4) Remove the cuvette from the chamber and unscrew the lid.

5) Empty the cuvette completely.

6) Mix the sample water thoroughly, rinse the cuvette just used with
sample water and then fill it with 10 ml of the sample water.

7) Replace the lid on the cuvette and insert it in the sampling chamber of
4 the PrimeLab. Align the arrow on the front of the cuvette. Press TEST.

8) The determined result is immediately displayed. Press the "OK" button

Note(s)
-> To get a more accurate indication of the measured value, a gravimetrical
determination of the water sample is necessary. Here, the water sample is
filtered and the residue evaporated at about 100 degrees and weighed to
the dried residue.

-> Make sure that the temperature of the water sample for measurement is
equal to the temperature of the water sample with extraction of the water
sample, otherwise measuring errors can occur.

-> In the best case do the measurement of the water sample immediately after
extraction of the water sample. Otherwise, keep seven days in a closed
glass or plastic container at max. 4 desgrees.

-> Make sure that the sample contains no air bubbles in the measurement.
If this is the case, remove the air bubbles by tapping it with the cuvette.

to initiate a test! The following is just a description of the test steps and does not show how
the PrimeLab works in general.

(59) Turbidity 20 - 1000 FAU

-> (*) conversion to FTU (same as FAU)

-> FAU stands for Formazin Absorption Units, different from the NTU
(nephelometric) method.

-> The measurement should be conducted immediately after sampling.

-> Air bubbles will influence the measurement result.

-> Tinted water samples influence the result. In this case do not use distilled
water (step 1) but rather filtered sample water for the ZERO adjustment.

-> The Turbidity test measures the optical value of the sample which results
from the scattering and absorption of light particles.
The amount of turbidity depends on variables such as size, shape, colour
and the refractive nature of the particles.
This test is calibrated using Formazin Turbidity Standards and the readings
are in terms of FAU (Formazin Attenuation Units).
This test can be used for daily plant monitoring and 1 FAU is equivalent to 1
NTU (Nephelometric Turbidity Unit).
This test is not suitable for USEPA reporting purposes as the optical method
of measurement for FAU is very different than the NTU method.
However 1 NTU = 1 FTU = 1 FAU when traced to formazin primary
standards.

1) Mix the sample water thoroughly, rinse cuvette -to be used for the
following measurement- several times and fill it with exactly 10ml of the
sample water. For this, you HAVE TO use the 10ml pipette which came
with your Turbidity Kit.
2) Insert the cuvette WITHOUT LID into the PrimeLab sampling chamber.
Align the arrow on the front of the cuvette.
3) Place Turbidity-Adapter on top of the open cuvette which already is
placed into the PrimeLab measurement chamber.

4 4) Press TEST.
5) The determined result is immediately displayed. Press button 3 to
3 convert the result to different units (*). Press button 4 to save the result
4 together with date and time.

!!! If low values (<20 NTU) are expected, we recommend to let the water
sample (in the vial) rest for at least 05:00 minutes before presing TEST.
As an alternative, you can also continue to repeat measurement in steps of
01:00 minute. The lowest value displayed can be taken as a result.

(112) Turbidity-NTU 0.02 - 1000 (NTU) Adapter

-> (*) Conversion to FTU / FNU

-> Ensure that all parts are clean, dry and free of grease and the adapter
must be placed firmly until it stops.

-> As this Parameter uses an indirect light from above, it is essential to always
ensure the same level of water in each vial to be tested.
Exactly 10 ml of liquid need to be used which can be achieved by using the
pipette provided with each kit.
Please change or clean the tip of the pipette after each measurement/
calibration by using destilled water.

-> If your PrimeLab was delivered with activated ID 112 (means you have NOT
activated it afterwards), the device is already calibrated. You only have to
make a new calibration again if you feel that results obtained are inaccurate.
The calibration process is under SET - described > calibration.

-> The following factors affect the accuracy of the measurement result :

-> One of the following reasons can lead into receiving an error message:
“check adapter”
• Weak or empty batteries (please change)
• Dirty lense (adapter)
• Wrong adapter used for this measurement (there are different adapter for
different measurements, all looking the same)
• Adapter might not stay straight on PrimeLab
• Cuvette-hole (PrimeLab) might be dirty (check the two windows)
• Water sample might be too dark / not enough light can pass water sample
to reach the sensor

-> The turbidity measurement method ID 112 uses, is based on the


Nephelometric principle, which is also described in DIN EN ISO 7027.
