1 Operating instructions
1.1 Using this manual

 Make sure that the service manual and the operating manual are available in the latest versions. To do
so, compare the version numbers. The latest versions of the manuals can be downloaded from
www.eppendorf-support.com and www.eppendorf.com.
 Read the service manual before commencing work on the device.
 Read the chapters "Installation" and "Operation" of the operating manual.
 Observe the safety instructions in the operating manual.

2 Product description
2.1 Hardware description

The electronics of the Centrifuge 5424 R consist of the following modules: PCB Display/Operation and PCB
Control/Converter.

The mains input module including switch and overcurrent protection devices is located on the right side of
the housing.

After the centrifuge is switched on, the hardware name and the firmware version of the control are
displayed shortly. The firmware can be updated via the USB interface.

2.1.1 PCB Display/Operation

This module mainly consists of control and input elements and an LC display. Its components and functions
are as follows:

• Input and output of centrifugation parameters, temperature, rotational speed, and time.
• Input and output (display by symbols) of all additional settings, e. g. "key lock", "At Set" function, etc.

There are two methods for entering centrifugation parameters:

• By rotary knob: Rotational speed and time are entered by means of two mechanical encoders; the
temperature is entered via arrow keys.
• By keypad: All centrifugation parameters are entered via arrow keys.

Further control and input elements:

• Start/Stop key
• Open key
• Rpm/Rcf key
• Menu/Enter key
• Menu arrow key
• Fast temp key
• Short key

2.1.2 PCB Control/Converter

This module controls and regulates all processes within the centrifuge. It is divided into two functional
areas: "Centrifuge control" and "drive & compressor control".

Centrifuge control
• Processing of signals received from the rotational speed, rotor bowl temperature, and condenser
temperature sensors.
• Control of all run parameters.
• Control of the electronics and condenser fans.
• Control of the solenoid valve for temperature control.
• Control of the speaker.
• Control of the lid-latch motor and recognition of the switching conditions of the individual lid switches.
The centrifuge control controls, via the driver circuit, the 12 Volt DC gear motor which locks and
unlocks the lid latch on the right and left sides. The locking condition is evaluated by means of a reed
switch (lid open/closed) and two microswitches (locking condition: locked/unlocked).

Drive and compressor control


• Interface for centrifuge control via optical coupler
• Switching power supply unit for 5 Volts and 12 Volts (floating), as well as for 5 Volts and 15 Volts
(energized).
• Radio interference filter.
• PFC circuit for 100 V, 120 V and 230 V.
For compliance with the EN 61 000-3-2 harmonics directive the voltage versions are equipped with an
active power factor correction (PFC) circuit essentially consisting of IC N25 (UCC 3818D), MOSFET V63
and the L10/ L32/ L33 PFC choke, which ensures that the line current drawn is almost sinusoidal.
Therefore, sufficient power is available for operating the drive and cooling even in the 100 V version.
• Braking chopper with external brake resistor.
• Module for control via centrifuge motor.
The intermediate circuit voltage and the intermediate circuit current are monitored.
• Module for control via compressor.
• Control of the electronics fan and the condenser fan.
The electronics fan is operated at different rotational speeds, depending on the cooling air required to
cool the centrifuge motor and the PCB Control/Converter.
The condenser fan is speed controlled as well, depending on the condenser temperature.
• Hardware safety circuit for lid switch and motor temperature safety switch outside of the processor
control.

When you switch the centrifuge off the current centrifuge parameters are stored in the PCB Control/
Converter. When you switch the centrifuge on again you can continue to operate with the centrifugation
parameters stored before.

2.1.3 Drive and rotational speed

The drive motor used is a three-phase asynchronous motor.

Motor characteristics
• n = 15,000 rpm
• P = 200 Watts

The acceleration and the braking of the centrifuge motor are ramp-controlled.

The rotational speed of the motor is measured by a Hall sensor located on a separate printed circuit board
below the motor.

The rotational speed signal (one impulse per revolution) is used for rotational speed control and display.

2.1.4 Cooling

An NTC resistor embedded in a metal sleeve on the bottom of the motor chamber serves as motor chamber
temperature sensor.

The compressor is a rolling piston compressor which is operated at 55 Hz.

For optimum operation of the cooling system the condenser temperature is maintained in a range between
35 and 40 °C. For this purpose the rotational speed of the condenser fan is adjusted in 32 steps (step 32 is
the maximum rotational speed), depending on the condenser temperature.

The temperature is controlled via a bypass with a solenoid valve situated on the hot gas side.

The coolant is injected into the evaporator by a thermostatic injection valve.

2.1.5 USB interface

A USB interface is installed behind the cover on the left side of the device. The USB interface can be used
to update the firmware of the centrifuge if required (see Software update on p. 49).

3.1 User profile

The specialist entrusted with maintenance, repair or testing must meet the following prerequisites:
• Successful participation in service trainings with certification by Eppendorf AG for the product that is to
be maintained, repaired or tested.

4.1 Overview of operating controls

The Centrifuge 5424 / 5424 R is available in two versions: keypad or rotary knobs. This operating manual
generally describes how to operate the keypad version. However, it also applies to the rotary knob version.

Before using the Centrifuge 5424 / 5424 R for the first time, familiarize yourself with the operating controls
and the display.

5.1 Error messages

Error messages consist of a maximum of 3 parts:

Up to firmware version 1.9: In case of errors, only the main error (e.g. Err 6) is displayed. To display the
complete error message (e.g. Err 6_2_4), press the rpm/rcf key for > 6 seconds. The display switches to
customer service function 9 (refer to “customer service functions" > “Error number / Error cause”). The
complete error message is displayed there. Customer service function 9 can only be accessed when an
error message is active. The FC error number is only relevant for error 6_2_xx. It is only displayed for this
error.

The complete error number is also displayed in service function SE 07.

5.2.2 Activation and operation

Proceed as follows to activate and operate the service functions:

1. To activate the service functions, press the rpm/rcf key and simultaneously switch on the centrifuge
using the mains power switch. Keep the Rpm/Rcf key pressed until SE 1 appears in the display.
2. To select a service function, turn the time rotary knob or press the time+ / time- keys.
3. To activate a service function, press the Start/Stop key.
4. To exit a service function and return to the selection screen, press the short key.
5. Switch off centrifuge to exit the service functions.

5.2.3.1 SE 1 (Measurement of the acceleration and deceleration times)

After activating SE 1, a short display test is performed. All segments and symbols are displayed
simultaneously. After that, the display advances automatically to Display 1.

Display 1: Now you can start the measurement run:


1. Close the centrifuge lid.
2. Press the Start/Stop key to start the
measurement run.
The centrifuge accelerates to maximum speed.
Then the acceleration time (Display 2) is
alternately displayed with the current run data

A brief display test is conducted following activation: all segments and symbols are first displayed
individually and then simultaneously (display 1).
After a few seconds the display advances automatically to the key test (Display 2).

5.2.3.5 SE 5 (Cyclical endurance test)

the initial start (display 2).

The operating hours and the number of load changes can be deleted by performing a full
initialization (SE 2).

5.2.3.9 SE 9 (Testing the overspeed cut-off)

NOTICE! Damage to device caused by excessive speed.


The centrifuge should automatically switch off when the cut-off speed of the inserted rotor has
been reached.

 Find out more information about the maximum speed of the inserted rotor.
 Observe the check.
 If the centrifuge does not automatically switch off, disconnect the centrifuge from the
power supply.

The inspection must be carried out with rotor!

Display: 1. Close the centrifuge lid.


2. Press the Start/Stop key to start the test run.
The centrifuge accelerates to cut-off speed and
must switch off in conjunction with Error 7.

Up to firmware version 1.9, the display advances automatically every second. From firmware
version 2.0 upwards, the display can be advanced manually using the start/stop key.

5.2.3.11 SE 11 (Testing the compressor and the solenoid valve)


The service function SE 11 has three displays. You can use Display 2 to test the proper functioning of the
compressor and the solenoid valve.
To continue switching the displays,
 Press the Menu/Enter key.

Display 1:

Display 2: • b: Compressor ready for operation (0 = not


ready, 1 = ready)
• c: Compressor status (0 = switched off,

5.2.3.13 SE 13 (Interface check)

Observe the descriptions of the interface check (see Interface test on p. 67) and "CefuComTest"
software on the www.eppendorf-support.com website. A password is required to access the
website.
Both devices need to be switched on in order to connect the centrifuge and the PC/laptop
using the USB cable.

Display 1: 1. Start the "CefuComTest" program on your


laptop/PC.
2. In the "CefuComTest" program, select the COM
port the centrifuge is connected to. Click
START.
3. Activate the interface check (SE 13) on the
centrifuge.
4. Press the start/stop key.
Data will be sent and received ("0x5A" will be
displayed in the "CefuComTest" program).
After the check has been completed successfully,
the display switches to display 2:
Display 2: 1. Click on STOP in the "CefuComTest" program.
2. Exit the service function.
3. Disconnect the USB connection.

"ERROR" will be displayed if the check was unsuccessful.


1. Check if the correct COM port is set in the "CefuComTest" program.
2. Check to see if the centrifuge and PC/laptop were switched on when the USB connection
was made.

5.2.3.14 SE 14 (Enter serial number of device)

It is possible to enter the serial number from firmware version 2.0 upwards.
The serial number has 12 characters and cannot be shown on the display. Therefore, the serial
number is entered in 2 parts

Display 1: Enter serial number part 1:


• bbbbbb: Part 1 of the serial number
Key functions:
• time+ / time- keys or time rotary knob: Select
position of the serial number.
• speed+ / speed- keys or speed rotary knob: Set
value of the serial number digit.
• start/stop key: Switch forward to part 2.

5.3 Customer service functions


5.3.1 Activation and operation

Proceed as follows to activate and operate the customer service functions:

1. To activate the customer service functions, press and hold the Rpm/Rcf key for more than 6 seconds.
2. To move on to the next display, press the Rpm/Rcf key briefly.
3. To exit the customer service functions, press and hold the Rpm/Rcf key for more than 6 seconds.
The customer service functions can also be selected when the centrifuge is running.
The functions of the Start/Stop and Open keys remain unchanged while the customer service
functions are activated. The centrifuge can be started and stopped. The lid can be closed and
opened.

5.3.2 Lid latch

6.1.10 Motor and motor suspension

NOTICE! Damage to the compressor and refrigerating system due to tilting the device.
The compressor and refrigerating system can become damaged if the device is switched on
when it is tilted or if the device is switched on right after it was tilted.

 Leave the device in the normal position for at least 4 hours before switching it on.
 Never place the device upside down.

Prerequisites
The motor can be changed without disassembling the housing. The centrifuge lid and the upper part of the
housing do not need to be removed.

When mounting the motor ensure that the motor sealing is completely in the groove of the
motor plate sleeve.
The sandpaper disks serve as an anti-twist lock. When mounting the motor, the nuts on the
sides with the sandpaper disks should be tightened last to prevent the rubber sleeve from
being twisted.

6.1.11 Brake resistor

Prerequisites
The centrifuge lid and the upper part of housing have been removed.

The desoldering fuse on the brake resistor may not be resoldered! If an error occurs, the
behavior of the fuse cannot be predicted if the type and quantity of the solder, and the
soldering temperature, are not an exact match.

 The entire brake resistor always needs to be replaced if the soldering fuse is open.

7 Alignment/adjustment
7.1 General adjustments

No adjustment of individual assemblies is required. Pay attention to the correct position of the cables and
free movement of the moving parts and assemblies. Make sure that the individual parts and assemblies
match.

The speed, temperature and time parameters as well as the safety arrangements have prescribed control
installations and thresholds that cannot be changed. It is not possible to adjust these parameters and
functions.

From firmware version 2.0 upwards, the serial number of the device must be entered in service function SE
14 after replacing the PCB Control/converter.

7.2 Adjustment of the reed switch

If the reed switch is mounted too low the lid latch may not be started when the centrifuge lid is
closed, resulting in the centrifuge lid not being locked.
If the reed switch is mounted too high the latch hooks may not reach the centrifuge lid,
resulting in the centrifuge lid not being locked.

Proceed as follows to adjust the reed switch:

 Mount the reed switch rather in the middle.


 Check the proper functioning of the lid latch with the housing and the centrifuge lid being mounted.
Correct the mounting position of the reed switch if required to ensure the proper functioning of the lid
latch.

8.1 Firmware

From firmware version 2.0 upwards, the serial number of the device must be entered in the service function
SE 14 after replacing the PCB Control/Converter.

8.2 Software update

In some cases, the software/firmware version of the PCB(s) does not match the current status.

 After replacing the printed circuit board(s) check the software version and update it, if
required.

The centrifuge firmware can be updated using a USB interface and a laptop/PC.

The interface is on the left-hand side of the device, behind the cover.

To ensure communication between the centrifuge and your laptop/PC you must install the
"CefuProgrammer" software (from version 1.1 on) and the drivers required on your laptop/PC.

The CefuProgrammer, driver, and instructions can be downloaded from the www.eppendorf-support.com
website (password required).

The current version of the firmware can also be downloaded from the www.eppendorf-support.com
website.

 Read the installation and operating instructions for the CefuProgrammer and the firmware
carefully before downloading, installing or updating the software!

10.1 Cleaning, disinfection, decontamination

 Switch off the device and disconnect the power plug before starting cleaning or
disinfection work.
 Do not allow any liquids to penetrate the inside of the housing.
 Do not spray clean/spray disinfect the housing.
 Only plug the device back in if it is completely dry, both inside and outside.

AM 01 Cleaning the device.

 Clean the centrifuge. Cleaning is described in the operating manual and in the
supplement sheets.
AM 02 Cleaning the condensation water tray and the hose.
1. Empty the condensation water tray and clean it.
2. Check that the tube is unobstructed.
3. Carefully blow the tube out with compressed air, if required.
AM 03 Checking the display.

 Check the display using the corresponding service function.


Pay attention to the following:
• Defective pixels
• Background lighting
 Replace display, if required
AM 04 Checking the keypad.

 Check the keypad using the corresponding service function. Press all of the keys.
 Check the tactility. The pressure on all of the keys must feel the same.
 Replace keypad, if required.
AM 05 Checking the housing

 Clean the housing. Cleaning is described in the operating manual and in the supplement
sheets.
 Check the housing for cracks. Liquid may enter the centrifuge through cracks.
 Check the housing for any damage that endangers safety.
 Replace the housing, if required.
AM 05 Checking stability

 Check the device feet for stability.


 If cracks have developed on the device feet, replace them.
AM 06 Checking the rotor bowl.
Cleaning of the rotor bowl is described in the operating manual.

 Inspect the rotor bowl for damage.


 Check the plastic rotor bowl for cracks.
 Clean the metal rotor bowl. The rotor bowl is made from stainless steel or has a special
coating.
 Check the rotor bowl for glass breakage and clean, if required.
 If samples with a high concentration of sodium chloride (NaCl) are used, spots may
appear which look like rust deposits. Remove any rust deposits with a polishing paste.
Rust deposits occur if the cleaning instructions recommended in the operating manual
were not followed.
 Replace the damaged rotor bowl.

AM 07 Cleaning and greasing the rotor bowl seal.


A description of the cleaning and care procedure can be found in the operating manual and
supplement sheets.
 Check the seal of the rotor bowl.
If the seal is defective, the lid will not close properly. More ambient air enters the rotor bowl.
The humidity entering the rotor bowl results in increased ice formation in the rotor bowl.
The cooling capacity of the centrifuge decreases.
Pay attention to the following:
• Seal with cracks
• Deformed seal
• Hardened seals
 Replace the damaged seal.
 Clean the seal of the rotor bowl.
 Maintain the seal of the rotor bowl, e. g. rub the seal with glycerin or grease.
AM 08 Checking the lid latch.

 Check the reed switch using the corresponding service function.


 Check the reed switch manually using a magnet.

Cracks and damage are caused by:
• Incorrectly sealed sealing clamp on the aerosol-tight caps.
• Tubes and jammed parts in the rotor bowl during centrifugation.

If the shock mounts for motor are mounted horizontally, they are exposed to shearing forces
and the weight of the loaded rotor. The shock mounts may develop cracks or may become
deformed.
 Check the shock mounts for motor.
Pay attention to the following:
• Cracks
• Damage
• Deformation
 Replace the shock mounts for motor every two years.
 Only replace shock mounts for motor as a full set (3 pcs.).
AM 15 Checking the imbalance detection.
1. Adjust the imbalance switch as described in the Alignment/Adjustment chapter of the
service manual.
2. Using the corresponding customer service function, check whether the tolerance is
observed for the adjustment.
AM 16 Checking motor fan and device fan.

 Use a cleaning brush or compressed air to remove any dust and contamination. Remove
contamination with an industrial vacuum.
 Listen for running noises.
 Use the corresponding service function to select the different program steps for testing.
 If the centrifuge is used frequently, replace the fan as a precautionary measure.

 Check all ventilation slots and openings. Clean them if required.

2. Load the rotor with the maximum number of filled tubes.
3. Only refrigerated centrifuges: Start run by pressing the fast temp key. 6°C should be

PS 10 Compulsory testing in Germany (BGR 500)
The accident prevention regulation BGR 500 (formerly VBG 7z) is valid in Germany.
Centrifuges with a rated power of > 500 W or a kinetic energy of > 10,000 Nm are subject to
testing.
The following Eppendorf AG centrifuges are subject to testing: Centrifuge 5804, 5804 R,
5810, 5810 R, 5810 RA, 5920 R.
These centrifuges must be tested once a year in their operating state and at least every 3
years in a dismantled state.
Rotors with a kinetic energy of ≤ 10,000 Nm are not subject to testing. Rotors that are used
in centrifuges subject to testing must be tested independent of their power output.
The owner of the centrifuge is obliged to document and keep the check results.

10.4 Device tests for centrifuges


10.4.1 Speed check

Measuring device
• Digital hand tachometer suitable for contactless measurement, or stroboscope
• Measurement range: 1 to ≥ 30,000 rpm
• Accuracy: ≤ 0.1%
1. Install the rotor.
2. Fully load the rotor with filled tubes.
3. Mark the rotor hub with a reflector (measuring device accessory).
4. Close the centrifuge lid.
5. Start the centrifuge with the desired speed. Let the centrifuge run until the speed has stabilized.
6. Measure the speed via the monitoring glass in the centrifuge lid using the operating manual of the
measuring device.
7. Note the measuring result.
8. If required, repeat steps 5 to 7 for other rotational speeds.

A recommendation on the rotational speeds to be tested, and their tolerances, can be found in the technical
data or OQ of the corresponding centrifuge.

10.4.2 Temperature check

Ambient temperature: 23°C ± 2°C


Measuring device
• Thermometer with a temperature sensor and a low internal heat capacity, e.g. thermocouple, max. Ø
0.5 mm)
• Measurement range: 0 °C to ≥ 100 °C
• Accuracy: ≤ 0.1°C
Auxiliary equipment

• Tubes that fit the rotor, with min. volume of 1.5 mL

10.4.2.1 Refrigerated centrifuges

1. Install the rotor and load it symmetrically with tubes.


2. Fill the tubes with ethylene glycol.
3. Set the maximum rotational speed.
4. Set the minimum temperature that can be achieved (depends on the rotor).
5. Precool the centrifuge and rotor with tubes and with adapters, if required, using fast temp (refer to the
operating manual for the centrifuge).
6. Start the centrifuge with the previously set max. speed and min. temperature.
7. Wait for 60 min. Then stop the centrifuge.
8. Measure the temperature in the center of the tubes immediately after opening.
9. Compare the sample temperature with the temperature displayed by the centrifuge.

10.4.2.2 Non-refrigerated centrifuges

For non-refrigerated centrifuges, sample heating is checked against the ambient temperature.

1. Install the rotor and load it symmetrically with tubes.


2. Fill the tubes with ethylene glycol.
3. Set the maximum rotational speed.
4. Start the centrifuge and let it run for at least 45 min.
5. Measure the ambient temperature.
6. Stop the centrifuge and measure the temperature in the center of the tubes immediately after opening.
7. Compare the ambient temperature and sample temperature.

A recommendation on the temperatures to be tested, and their tolerances, can be found in the technical
data or OQ of the corresponding centrifuge.

10.4.3 Timer check

Measuring device
• Stopwatch
• Accuracy: ≤ 0.1 seconds
1. Set the run time to continuous operation.
2. Start the centrifuge and stopwatch at the same time.
3. Allow the centrifuge to run at least 5 min.
4. Compare the time displayed by the centrifuge with the time measured by the stopwatch.

10.4.4 Electrical safety check

Notes concerning the measuring procedure:


• The electrical safety check may only be carried out by a suitably trained electrician.
• After repairs or after work on the electrical system of the centrifuge (incl. protective earth conductor
system) the electrical safety must be tested.
• The electrical safety must be checked at regular intervals (e.g. every 12 months).
• Measure on a grounded, metallic point that is not insulated and can be touched from the outside (e.g.
housing screws).
• The leakage current or resistance of the PE conductor cannot be measured at the centrifuge lid, rotor, or
motor shaft.
• Never measure at any interfaces that may be present. The centrifuge electronics could become
damaged.
• Due to the internal electrical design of the centrifuge, the insulation resistance cannot be measured
correctly.
• Due to the internal electrical design of the centrifuge, the equivalent leakage current cannot be
measured correctly.
→ Always measure the real leakage current (direct measurement or differential current measurement).

10.5 Interface test

The function of the USB interface can be tested using the "CefuComTest" program and a laptop/PC.

To test the interface, the "CefuComTest" software and corresponding driver must be installed on your
laptop/PC. The "CefuComTest", driver, and instructions can be downloaded from the Support webpage:
www.eppendorf-support.com (password required).

Read the installation and operating instructions for the "CefuComTest" carefully before
starting downloading, installation or the interface test!
For the interface test, the corresponding service function must be activated. Observe the
description of the service function in chapter 5.
