The Goal
1

 To familiarize students with the fundamental and basic concept of

microscopes and their significant in biology.

At the end of the lecture lab

 Student will:
 Understanding microscope .
 The two main types of microscope.
 Parts of microscope.
 Usage of microscope.
Microscope
2

 A microscope is a scientific
instrument/tool that magnifies
and allows the observation of tiny
objects or details that are not
visible to the naked eye.
 The first compound microscope
was invented in 1595 by he

Types of Microscope
4

1.Light microscope: Use light to magnify small objects widely

used in biology.
2.Electron microscope: Use electron for magnification. There are
two types: a. Transmission Electron Microscope (TEM). B.
Scanning Electron Microscope (SEM).
3.Confocal microscope: Use laser beam.
4.Fluorescence microscope: Use fluorescence to visualize
structure.

1. Eyepiece (Ocular Lens): The lens at the top of the microscope that you look through.
Typically, it magnifies the image 10x.
2. Objective Lenses: These are multiple lenses attached to a rotating nosepiece. They provide
different levels of magnification (usually 4x, 10x, 40x, and 100x) by varying their focal
lengths.
3. Stage: The platform where the specimen or slide is placed for observation.
4. Specimen Holder/Stage Clips: Clips or mechanical holders used to secure the slide in place
on the stage.
5. Coarse Focus Knob: A larger knob used for initial rough focusing by moving the stage up
and down rapidly. It is used with low-power objectives (4x and 10x).
6. Fine Focus Knob: A smaller knob used for precise focusing to bring the specimen into sharp
detail. It is used with high-power objectives (40x and 100x).

7. Base: The bottom part of the microscope that provides stability and houses.
8. Arm: The curved part of the microscope that connects the tube to the base and provides
support for carrying the microscope.
9. Diaphragm: An adjustable disk under the stage that controls the amount of light entering the
condenser.
10.Condenser Lens: Located under the stage, it concentrates and directs light through the
specimen.
11.Source of Light
12.Light intensity control: to high or low the light intensity.
