ABOUT PHARMACEUTICAL INDUSTRY –

The pharmaceutical industry discovers, develops, produces, and markets pharmaceutical drugs for the
use as medications to be administered to patients (or self-administered), with the aim to cure and
prevent diseases, or alleviate symptoms. Pharmaceutical companies may deal in generic or brand
medications and medical devices.The main goal of this industry is to provide drugs to maintain health
or prevent infections and cure diseases affecting the global population.They are subject to a variety of
laws and regulations that govern the patenting, testing, safety, efficacy using drug testing and
marketing of drugs. The pharmaceutical industry can be viewed as a complex system of processes,
operations, and organizations involved in the discovery, development, and manufacturing of drugs.

The modern era of the pharmaceutical industry of isolation and purification of compounds,chemical
synthesis and computer aided drug design is considered to have begun in the 19 th century, thousand of
years after intuition and trial and error led humans to belive that plants,animals, and minerals
containted medicinal properties. The unification of research in the 20 th Century in fields such as
chemistry and physiology increased the understanding of basic drug discovery process. The continual
evolution and advancement of the pharmaceutical industry is fundamental in the control and
eleimation of disease around the world.

2-Raw material department -


A raw material warehouse is a facility that stores components or materials that will subsequently be
used on production lines to manufacture products.
In this industry we saw raw material department & all the raw materials are arranged in alphabetical
order. Raw materials are input substances used in both chemical synthesis and processing and include
buffers, cleaning agents, common solvents and commonly used synthetic starting materials such as
amino acids. Excipients are substances which are used as ingredients in pharmaceutical formulations
such as tablets and capsules..

PRODUCTION AREA –
A production department in the pharmaceutical environment includes various needs: distribution,
welding, packaging or cutting. Automating the pharmaceutical production line increases productivity
and improves the profitability of the company using it.
Pharmaceutical processing is the process of drug manufacturing and can be broken down into a range
of unit operations, such as blending, granulation, milling, coating, tablet pressing, filling and others.
The Pharmaceutical manufacturing process has precise requirements and manufacturing guidelines in
terms of quality.
[A]-Tablet Manufacturing –
A tablet (also known as a pill) is a pharmaceutical oral dosage form (oral solid dosage, or OSD) or
solid unit dosage form. Tablets may be defined as the solid unit dosage form of medication with
suitable excipients. It comprises a mixture of active substances and excipients, usually in powder form,
that are pressed or compacted into a solid dose. The main advantages of tablets are that they ensure a
consistent dose of medicine that is easy to consume.
Tablets are prepared either by moulding or by compression. The excipients can include diluents,
binders or granulating agents, glidants (flow aids) and lubricants to ensure efficient tableting;
disintegrants to promote tablet break-up in the digestive tract; sweeteners or flavours to enhance taste;
and pigments to make the tablets visually attractive or aid in visual identification of an unknown tablet.
A polymer coating is often applied to make the tablet smoother and easier to swallow, to control the
release rate of the active ingredient, to make it more resistant to the environment (extending its shelf

life), or to enhance the tablet's appearance.

Advantages –

1) Tablets are unit dosage form and offer the greatest capabilities of all oral dosage form for the
greatest dose precision and the least content variability.
2) They are easiest and cheapest to package and strip.

3) Low in cost.
4) Lighter and compact .
Disadvantages –

1) Difficult to swallow in case of children and unconscious patients.


2) Some drugs resist compression into dense compacts, owing to amorphous nature, low density
character.
3) Drugs with poor wetting, slow dissolution properties, optimum absorption high in GIT may be
difficult to formulate or manufacture as a tablet that will still provide adequate or full drug
bioavailability.

❖ Double cone blender -Double Cone Blender is a mechanical mixing equipment that is
extensively used in the pharmaceutical, food, chemical, and cosmetic industries to produce
homogeneous mixtures of dry powders and granules for tablets and capsules. The unique principle
of the double cone blender is based on the mechanical mixing that occurs due to tumbling motion.
The double-cone blender is constructed by welding two cone-shaped sections at their bases to a
central cylindrical section.

❖ Mass mixer -Mass Mixer Machine refers to any type of mixing device that provides for the
operation of kneading, bowl turning & shaker operations at high speed. The main advantage of this
type of equipment is that they help in speedy and convenient blending of large quantities of
ingredients, making the mixing process of many batches easier & less time consuming. They offer
maximum speed in mixing several batches at a time. Most mass mixers are operated by using a
screw pump. The mass mixer machine is basically just mixing a single mass with the help of a
rotating stirrer that is in vertical position inside the container & hence have only a single speed,
easy rotation speed.

❖ Hammer Mill - A Hammer Mill machine is widely used in the Pharmaceuticals and Food
industries to cutting Materials into small fragments or small pieces (Also called Size Reduction). The
main component of a hammer mill is a row of hammers that are hinged on a central shaft and
contained in a strong metal container. Size reduction is achieved by Impact .

❖ Tray dryer -The principle of a tray dryer is based on the process of convection drying.
Convection drying involves the use of hot air to remove moisture from the material being dried.
The hot air is circulated throughout the drying chamber, and as it passes over the trays, it removes

moisture from the materials on the trays. The temperature and humidity inside the chamber are
controlled to ensure that the materials are dried at the appropriate rate. in short; hot air is
continuously calculated inside the chamber, and forced convection heating takes place to remove
moisture from the solid spread in trays.

❖ Multi Station 20 rotary press machine - Multi-station press is a mechanical device that
unlike the single punch tablet press has several stations which rotate to compress granules/powder
mixture to a uniform size, shape, and weight of tablets with high productivity in a short time. It
was developed to raise the productivity of tablets.

Rotary tablet press is a mechanical device that unlike the single punch tablet press has several tooling
station which rotates to compress granules/powder mixture into tablets of uniform size, shape
(depending on the punch design) and uniform weight. It was developed to increase the output of
tablets.

In rotary tablet press, the compaction force on the fill material is exerted by both the upper and lower
punches leaving the powder granules to be compressed in the middle. This is known as accordion type
of compression. The capacity of a rotary tablet press is determined by the rotation speed of the turrent
and the number of stations on the press.

❖ Coating pan - Coating pan is utilized to create a coat around the pellets with the help of some
aqueous or organic film. The components of this device include a semi ellipsoidal pan fitted with a
circular cover to facilitate the charging and discharging of tablets. This pan is rotated with the help
of a motor and a gear.

➢ Capsule manufacturing –

Capsules are one of the essential components of the pharmaceutical sector. They are mainly used to
hold dry powder or little pellets of medication and are made using specific machinery and techniques.
The capsule's production method entails pharmaceutically active ingredients, additives, and a cover.
Every pharmaceutical business understands capsule manufacturing takes time, and the reason is the
critical measures.

Did you know?

Capsules are solid forms of medication in which contents are encased in either a soft or hard,
dissolving container or shells made from appropriate gelatins. Capsules have the benefit of being
palatable, readily ingested, and widely available in large numbers.

Capsules are classified into two categories based on the amount of gelatin present.

• Hard Gelatin Capsule& Soft Gelatin Capsule .


The fundamental difference between the two is that hard gelatin capsules are less plasticized than soft
gelatin capsules. In addition, hard gelatin capsules have two parts, the "cap" and "body". Hard gelatin
is widely used and filled with powder drugs or medicine pellets.

The Dipping Method used to manufacture hard gelatin capsules involves steps like Dipping, Rotation,
Drying, Striping, Trimming, and Joining.

Hard gelatin capsules, unlike the soft form, are made of two parts, the body and the cap, each of a
different colour. This form of capsule holds dry ingredients in powder form. The body is first filled
with the mix of active ingredients and any excipients used, and then closed with the cap using either a
manual or automatic press machine.

Soft Gelatin Capsules also called soft gels, soft gelatin capsules are a case produced from a single
piece of gelatin, rather than two halves attached together. They are used for solutions not based on
water, as this would dissolve the gelatin, but for oil-based solutions.

The manufacture of pharmaceuticals, encapsulation refers to a range of dosage forms—techniques
used to enclose medicines—in a relatively stable shell known as a capsule, allowing them to, for
example, be taken orally or be used as suppositories. The two main types of capsules are:

• Hard-shelled capsules, which contain dry, powdered ingredients or miniature pellets made by e.g.,
processes of extrusion or spherization. These are made in two-halves: a smaller-diameter “body”
that is filled and then sealed using a larger-diameter “cap”.
• Soft-shelled capsules, primarily used for oils and for active ingredients that are dissolved or
suspended in oil.

Both of these classes of capsules are made from aqueous solutions of gelling agents, such as animal
protein (mainly gelatin) or plant polysaccharides or their derivatives (such as carrageenan’s and
modified forms of starch and cellulose). Other ingredients can be added to the gelling agent solution
including plasticizers such as glycerin or sorbitol to decrease the capsule's hardness, coloring agents,
preservatives, disintegrants, lubricants and surface treatment .

❖ Syrup manufacturing –

Syrup is a medicine in the form of a thick liquid containing a sugar solution. The cough medicine is
supplied as a thick, sweet syrup. The thick syrup is sweetened with a solution of sugar. Syrup is a
medicine in the form of a thick liquid containing a sugar solution .Syrups is a concentrated solution of
sugar in water or other aqueous solutions. Medicinal syrup is a nearly saturated aqueous solution in
which the medicinal substance or drug is dissolved. Basically, it is an oral suspension in liquid form.
Medical syrup or medicinal syrup is actually used as a carrier for drugs.

Types of Syrups:

1. Simple syrup: Simple syrup is made by adding sucrose into purified water

2. Medicated syrup: Medicated syrup contains API (Active Pharmaceutical ingredients) with their
excipients.

3. Flavored syrup: Flavored syrup mainly contains aromatic or flavored excipients are called Flavored
syrup.

• Advantages of Syrup:
✓ Simple syrup is used as a preservative because it reduces bacterial and fungal growth due to high
osmotic pressure.
✓ Sugar syrup also acts as an antioxidant by hydrolyzing sugar into sugar laevulose and dextrose.

• Disadvantage of Syrup:

✓ Delay onset of action .


✓ Not suitable for emergency.
✓ Not convenient with gastrointestinal disorders.

❖ Liquid filling machine - A filling machine, or liquid filling machine, is a device for filling
containers, such as bottles or jars, with liquid in a repeatable and productive way. –

Filling machines can be used for any quantity of production, from just a few jars of liquid up to
fullscale production of thousands – or tens of thousands – of bottles.

Liquid filling machines will typically draw liquid from a large container such as an IBC tank, a hopper
or similar, and dispense a predetermined quantity of liquid into bottles, jars or other containers via a
nozzle.

Smaller scale liquid filling machines are often either manually operated (via a hand operated lever), or
semi-automatic; powered by compressed air, electricity, or a combination of the two.

Bottle filling machines are used across a wide range of industries, from pharmaceuticals and cosmetics
to solvents and paints, with everything included between.

❖ Liquid rubber cap sealing machine - Cap Sealing Machine Cap Sealing Machine is used for
sealing the cap of plastic containers and plastic bottles. The machines work while being place in a
table at a fixed height and the bottle moves up and down via the use of a table and thus, sealing is
done.

❖ Ointment manufacturing–

An oil-based topical formulation with a semi-solid texture and a greasy appearance that can be applied
to the skin is called an ointment. As per the ointment meaning, the therapeutic substances are dispersed
in the medium. The medium generally has 80% oil and 20% water. Ointments are preparations for
external use, intended for application to the skin. Typically, they have an oily or greasy consistency
and can appear “stiff” as they are applied to the skin.

Ointments. Of all skin products, ointments contain the most oil. They stay on top of your skin rather
than being absorbed right away, which offers more protection against moisture loss and the elements,
like cold or dry air. Common ingredients found in ointments include mineral oil and petroleum.

There are three major types of ointment bases: oleaginous (greasy) bases, which are water repellent;
water-absorbing bases, which are greasy but allow absorption of water, and water-miscible bases
(vanishing creams or water-soluble bases).

QUALITY ASSURANCE AND QUALITY CONTROL –

➢ Quality Assurance (QA) is the term used in both manufacturing and service industries to describe
the systematic efforts taken to assure that the product delivered to customer meet with the
contractual and other agreed upon performance, design, reliability and maintainability expectations
of the customer. The core purpose of Quality Assurance is to prevent mistakes and defects in the
development and production of both manufactured products, such as automobiles and shoes, and
delivering services, such as automotive repair and athletic shoes design. Assuring quality and
therefore avoiding problems and delays when delivering products or services to customers is what
ISO 9000 defines as that “part of quality management focused on providing confidence that quality
requirements will be full filled”.

➢ QUALITY CONTROL (QC) is a process by which entities review the quality of all factors
involved in production ISO 9000 defines quality control as “a part of quality management focused
on fulfilling quality requirements”. This approach places emphasis on three aspects (enshrined in
standards such as ISO 9001) . Quality control (QC) is a procedure or set of procedures intended to
ensure that a manufactured product or performed services adheres to a defined set of quality

➢ U.V Spectrophotometer -UV-Vis spectroscopy is based on the interaction between light and
matter. When light passes through or is absorbed by a molecule, it can cause the molecule to
vibrate. The wavelength of light that is most strongly absorbed by a molecule is called the
absorption maximum. UV-Visible spectroscopy exploits the wave-like nature of electrons and its
interactions with electromagnetic radiation. UV-Vis spectroscopic instruments are used to identify,
characterize, and quantify an extraordinary wide range of molecular compound .

➢ HPLC [ High Performance Liquid Chromatography]-High-performance liquid


chromatography or commonly known as HPLC, is an analytical technique used to separate,
identify or quantify each component in a mixture The mixture is separated using the basic principle
of column chromatography and then identified and quantified by spectroscopy. HPLC is thus
basically a highly improved form of column liquid chromatography. Instead of a solvent being
allowed to drip through a column under gravity, it is forced through under high pressures of up to
400 atmospheres.

➢ p.h meter - Potentiometric pH meters measure the voltage between two electrodes and display the
result converted into the corresponding pH value. They comprise a simple electronic amplifier and
a pair of electrodes, or alternatively a combination electrode, and some form of display calibrated
in pH units.It usually has a glass electrode and a reference electrode, or a combination electrode.
The electrodes, or probes, are inserted into the solution to be tested.[8] pH meters may also be
based on the antimony electrode (typically used for rough conditions) or the quinhydrone
electrode.

➢ Friability apparatus –Tablet friability testing involves weighing the sample of tablets and then
placing them into a rotating drum. The drum is then rotated 100 times. The sample is then
reweighed to find the % weight loss. Company's Digital Friability Test Apparatus is a precision
instrument for measuring obsession strength of the tablets. The Friability Test Apparatus is
designed as per IP/USP standards. The unit is equipped with two transparent acrylic drums which
rotate at a speed of 25.

➢ Disintegration apparatus -Disintegration testers work by raising and lowering a 'basket' in


and out of the test medium, for a set period of time, to see if the tablet dissolves or not. The test

Pharmaceutical packaging (or drug packaging) is the packages and the packaging processes for
pharmaceutical preparations. It involves all of the operations from production through drug
distribution channels to the end consumer. maintains drug quality. It protects the product from physical
damage as well as biological degradation. Some sensitive drugs require protection from light and
water as they have sensitive substances. The packaging for pharmaceuticals also needs to disseminate
important information. There must be proper and clear labelling—correct information needs to be
disseminated. Product packaging must protect the medicines from contamination and all external
influences that may alter the drugs’ property .

➢ Types of Packaging –

1-Blister Packaging -Blister pack is the most common pharmaceutical packaging used to hold solid
medicines in place. These are pre-formed foil, paper, or plastic packs. Blister packs have a pocket or
cavity made from thermoformed plastic. At its backside, there is a paperboard, aluminium foil, or
plastic film seal that can be easily punctured by hand. Notably, this seal includes all the important
information related to the drug.

Alu Alu blister packaging is a specific type of blister packaging commonly used in the pharmaceutical
industry. It is known for its excellent protective properties and is specifically designed to provide a
high level of protection to sensitive pharmaceutical

2-Strip Packaging - It is a unit packing dosage and is specifically used to increase the dosage life as it
protects the content individually. The most significant difference between blister and strip packaging is
that strip doesn’t have thermo-formed cavities. Instead, the packaging is formed around the tablet.

3-Bottle Packaging -Bottles are commonly used for liquid pharmaceuticals as well as formed
tablets and capsules. Glass is most common for liquids because it is inert and has excellent barrier
properties.

Storage conditions. Stability of drugs depends on both environmental factors such as temperature, air,
light and humidity, and drug-related factors such as the active ingredient itself, the dosage form (tablet,
solution, etc.) and the manufacturing process. This practice prevents possible contamination from dirt,
the environment, or insects. All pharmaceutical products must be stored within their labeled
temperature zones. Some examples are: Store at 2°C - 8°C.

Proper storage of pharmaceutical products is important to ensure the identity, strength, quality, and
purity of the products are not affected. Storage areas:

• Must have the appropriate conditions of temperature, humidity, and light


• Must prevent unauthorized persons from entering
• Should be of sufficient capacity
• Should be clean and dry
• Should be free of accumulated waste and vermin
• Should protect materials and products from weather
• Should segregate quarantined, approved, and rejected materials.
