Preface
The use of optical techniques in experimental biology dates back over 300
years, and has advanced through a series of technological breakthroughs exem-
plified by the development of diffraction-limited microscopy at the end of the
nineteenth century. We are now, once again, at a time of explosive progress, and
the newly christened field of biophotonics is emerging from innovative research at
the interface of the physical, biological, medical, and engineering sciences. Bio-
photonics encompasses a broad range of techniques and methodologies developed
in areas as diverse as photophysics, photochemistry, optical spectroscopy, and
microscopy. It is thus highly interdisciplinary in nature. For example, an imaging
technique based on concepts and principles borrowed from physics, chemistry, and
engineering may be used to investigate a biological system at the level of a single
molecule on an engineered surface within a living cell or even within an animal.
Distinct from previous publications devoted to specific topics, such as imag-
ing or spectroscopy, this 2-volume work of Methods in Enzymology is the first to
attempt a comprehensive coverage of the broad field of biophotonics research. We
believe it will prove a valuable resource for researchers interested in developing
and applying photonic technologies to solve biological problems at all levels, from
single biomolecules to the living organism. The chapters are written by internation-
ally renowned researchers, and provide technically detailed coverage of the basic
principles of optical spectroscopy and microscopy with numerous applications of
specialized techniques and probes in biology, medicine, and biotechnology.
Given the interdisciplinary nature of biophotonics, we failed in our initial at-
tempt to classify the chapters within defined categories of technology, applications,
and biological systems. Nevertheless, chapters in the two volumes are roughly
equally divided among three main areas. Biophotonics, Part A (Volume 360) cov-
ers the basic principles and practice of biological spectroscopy and imaging mi-
croscopy, with supporting chapters on photophysics, optical design, and biological
and synthetic probes. Biophotonics, Part B (Volume 361) focuses on the develop-
ment and application of imaging technologies to understand the mechanisms un-
derlying biomolecular structure, function, and dynamics in diverse molecular and
cellular systems. It also illustrates how biophotonic technologies are being used to
power new breakthroughs in medical imaging and diagnosis and in biotechnology.
