Reticulocyte

RET ( 0.025~0.075×1012/L )
 Between Metarubricyte and Mature erythrocyte.
 Important indicator for Anemia treatment

NRBC -- Nucleated red blood cell

NRBC ( 0~0.02×109/L )

 There will be no NRBC in normal

human peripheral blood, except for
neonate and fetus.
 Because NRBC have cell nucleus
inside, it is always identified as
WBC.

Fluorescence

 Feature of fluorescence
 Lower intensity
 Longer wavelength Fluorescence
 Affect by temperature Excitation light

Fluorescence -- nuclear acid

Dye + Nuclear ACID Compound

 Key questions:
a) How does the dye get into cell ? --- Affect the combination of dye and
nuclear acid
b) How much is the nuclear acid? --- Affect the target quantity of the dye
c) The density of the chromatin? --- Affect the efficiency of the bonding
process
d) The concentrate of dye? --- Affect the intensity of the fluorescence
e) Higher temperature, lower fluorescence intensity
f) Reaction time
g) Other factors
