Title: Plan and Design – Paper Chromatography

Statement of Problem:

Jim bought four different brands of black permanent markers. He believes that all four
brands of the black markers contain the same dyes
Plan and design an experiment to determine if the black ink from the four different
brands of permanent markers contains the same dyes

Hypothesis: The four different permanent markers may contain the same dye proven by
chromatography.

Aim: To determine if four brands of permanent markers contain the same dyes, using
paper chromatography.

Materials and Apparatus:


-Beaker -Aluminum Foil
-Glass Rod -Ruler
-Chromatography Paper -4 brands of permanent marker
-Tape -Pencil
-Solvent(water)

Method:
1. With the help of a pencil draw a light horizontal pencil line 2 cm from the bottom of
the chromatography paper.
2. Using the first marker brand A place a dot onto the middle of the baseline.
3. Into the beaker pour a sallow amount of the solvent(water).
4. Attach the chromatography paper onto the glass rod and secure it properly with tape.
5. Suspend the chromatography paper into the beaker which contains the water and
ensure that the solvent level does not pass the baseline.
6. Let the solvent(water) levels rise up the chromatography paper until it reaches the top
for 15 minutes.
7. Take out the chromatography paper out of the beaker and allow it to air dry.
8. With the aid of a pencil mark the position which the solvent travelled.
9. Take measurements of the distance from the solvent front to the baseline.
10. Take measurements of the distance of each individual component from the baseline.
11. Calculate the Rf value for each component.
12. Repeat steps 1-12 using the other brands of permanent marker B, C and D
Variables:

Controlled Variable
1. amount of time
2. size of the dot

Manipulated Variable
The various types of brands that is permanent brand A,B,C,D

Responding Variable
1. Distance travelled by the variables
Assumptions and Precautions:
Assumption
All pigments in the ink are soluble in water

Precautions
Ensure that a pen is not used to draw the line on the filter paper, as ink is soluble in the
solvent and it will interfere with the chromatogram.
Ensure that the ink sample placed on the penciled line is small.

Expected Results:
If the chromatograms are the same, then the black markers are identical and the
hypothesis can be accepted. If the chromatograms are not identical then the hypothesis
will be denied.
