1) How do enzymes work? If you know a chemical reaction and want to use an enzyme to catalyze
this reaction but don’t have a known enzyme that does this what will you do? (15 points)
2) You have a peptide ligand, but you don’t know the receptor for this protein. Design and explain
a set of experiments you can use to identify the receptor for this known ligand. (Hint: use
purification, detection confirmation, etc. Techniques you learned in this course) (15 points)
3) You have a cell line that responds to a specific Growth Factor (possibly acting through receptor
tyrosine kinase) and a second cell line that does not respond to the same growth factor. What
can you do to show this growth factor acts through RTK. How does RTK signal in the cell? Can
you use this information to prove your growth factor is working through the RTK pathway?
Finally, can you find the possible RTK that interacts with your growth factor? (15 points)
4) What are G-proteins? Explain and describe how they play role in signal transduction. Gave two
examples of where these proteins are used in a biological system. (15 points)
5) Explain the roles of Lipids in detail, give examples for various lipids carrying different functions.

6) You are working with a protein, and there is a possibility that this protein is a glycoprotein. How
can you find if this protein is glycosylated? Identify which residues are glycosylated (explain in
detail)? (15 points)
7) After sequencing, you discovered that the protein you have contains an SH2 domain, a DNA
binding domain and a histone acetyltransferase (HAT) activity. Guess the possible role and
action mechanism of this protein, and propose experiments that you can do to test this. (15
points)
